Description
The Mayo Clinic PKD Center comprises a team of expert faculty engaged in basic research and providing comprehensive adult and pediatric PKD care. Our mission spans from fundamental discovery through translational analyses in model systems and human subjects to conducting clinical trials. We actively collaborate and share materials, resources, and expertise to advance both academic and industry-sponsored PKD research. Our primary goal is to foster breakthroughs in basic research and their application in the PKD clinic.
The Center focuses on three main areas: Genetics, Epigenetics, and Metabolomics (GEM), Model Systems, and Imaging. The GEM core specializes in genetic profiling of PKD patients to unravel disease origins and identify potential disease mechanisms and biomarkers. We oversee a Cystic Kidney and Liver Disease Biobank for the collection and dissemination of PKD patient materials. Additionally, we manage the PKD Foundation ADPKD Mutation Database, evaluating variants in the PKD1 and PKD2 genes for their clinical significance.
The Model Systems core develops and maintains PKD models in C. elegans, zebrafish, rodent, and pig for collaboration or sharing. We offer standardized protocols for zebrafish and murine preclinical testing to assess novel therapeutic efficacy. For high-throughput drug screening, we offer orthologous C. elegans and zebrafish models.
The Imaging core continuously advances protocols for preclinical and clinical imaging, including 3D ultrasound, CT, and quantitative MR, using state-of-the-art image processing techniques like radiomics, machine learning, and deep learning analysis. Standardized protocols are available to ensure accurate and automated analysis of organ volume, cyst number, and size, providing precise patient-specific image-based metrics.
In addition, the Center develops and tests novel technologies, such as base editing mediated gene therapy and RNA therapy in PKD. We also actively support aspiring researchers from undergraduates to junior faculty, offering opportunities such as Summer Undergraduate Research Fellowships funded through a R25 grant and Postdoctoral Fellowships.
